Fertilization triggers cytoplasmic movements in the frog egg that lead in mysterious ways to the stabilization of β‐catenin on the dorsal side of the embryo. The novel Huluwa (Hwa) transmembrane protein, identified in China, is translated specifically in the dorsal side, acting as an egg cytoplasmic determinant essential for β‐catenin stabilization. The Wnt signaling pathway requires macropinocytosis and the sequestration inside multivesicular bodies (MVBs, the precursors of endolysosomes) of Axin1 and Glycogen Synthase Kinase 3 (GSK3) that normally destroy β‐catenin. In (...) Xenopus, the Wnt‐like activity of GSK3 inhibitors and of Hwa mRNA can be blocked by brief treatment with inhibitors of membrane trafficking or lysosomes at the 32‐cell stage. In dorsal blastomeres, lysosomal cathepsin is activated and intriguing MVBs surrounded by electron dense vesicles are formed at the 64‐cell stage. We conclude that membrane trafficking and lysosomal activity are critically important for the earliest asymmetries in vertebrate embryonic development. (shrink)

On the basis of his acquaintance with theoretical elementary particle physics, and following the lead of Thomas Torrance, John Polkinghorne maintains that the data upon which a science is based, and the method by which it treats those data, must respect the idiosyncratic nature of the object with which the science is concerned. Polkinghorne calls this the "accommodation" (or "conformity") of a discipline to its object. The question then arises: What should we expect religious experience and theological method to be (...) like if they are accommodated to the idiosyncratic nature of God? Polkinghorne's methodological program is typical of postcritical positions in the theology-science dialogue in holding that the fiduciary element in theological method is simply a species of the fiduciary element that is a de facto part of all knowing—in other words, theological method does not differ in fundamental kind from the methods of the natural sciences. But this program may contain the seeds of an alienation of theological method from the transcendence of God similar to the double self-alienation of theology described by Michael Buckley in At the Origins of Modern Atheism. I contend that something like Bernard Lonergan's position on how the method of faith seeking understanding is related to the methods of the natural sciences is exactly the sort of thing that one should expect on the supposition of Polkinghorne's principle of accommodation, at least if the God who is the object of theological science is transcendent. The way in which the divine differs from all other objects ought to be disclosed or reflected in religious experience and theological method. Polkinghorne charts the course for an accommodated theology, but it seems to be Lonergan who is more intent on following it. (shrink)

BackgroundThe ARRIVE (Animal Research: Reporting of In Vivo Experiments) guidelines are widely endorsed but compliance is limited. We sought to determine whether journal-requested completion of an ARRIVE checklist improves full compliance with the guidelines.MethodsIn a randomised controlled trial, manuscripts reporting in vivo animal research submitted to PLOS ONE (March–June 2015) were randomly allocated to either requested completion of an ARRIVE checklist or current standard practice. Authors, academic editors, and peer reviewers were blinded to group allocation. Trained reviewers performed outcome adjudication (...) in duplicate by assessing manuscripts against an operationalised version of the ARRIVE guidelines that consists 108 items. Our primary outcome was the between-group differences in the proportion of manuscripts meeting all ARRIVE guideline checklist subitems.ResultsWe randomised 1689 manuscripts (control: n = 844, intervention: n = 845), of which 1269 were sent for peer review and 762 (control: n = 340; intervention: n = 332) accepted for publication. No manuscript in either group achieved full compliance with the ARRIVE checklist. Details of animal husbandry (ARRIVE subitem 9b) was the only subitem to show improvements in reporting, with the proportion of compliant manuscripts rising from 52.1 to 74.1% (X2 = 34.0, df = 1, p = 2.1 × 10−7) in the control and intervention groups, respectively.ConclusionsThese results suggest that altering the editorial process to include requests for a completed ARRIVE checklist is not enough to improve compliance with the ARRIVE guidelines. Other approaches, such as more stringent editorial policies or a targeted approach on key quality items, may promote improvements in reporting. (shrink)

Na primeira parte, explicita a dimensão antropológica e os pressupostos críticos de compreensão de uma tradição religiosa. Na segunda, recolhe contribuições importantes da tradição judaico-cristã (...) para a efetivação do diálogo interreligioso: a estrutura dialógica da revelação divina, a universalidade do amor de Deus, a promoção da vida humana como valor central, a necessária encarnação histórica da experiência de Deus e a dimensão sacramental da religião diante da centralidade teológica do Reino de Deus. Por fim, utiliza a categoria inreligionação, de Torres Queiruga, para mostrar as maravilhas do possível e necessário diálogo inter-religioso. ABSTRACT The author extracts from Christian experience perspectives associated with, and necessary to, the historical concretization of inter-religious dialogue. According to his thesis, basic elements of inter-religious dialogue are present in Jesus’ own religious experience. The starting point is the acknowledgment of the legitimacy of cultural and religious pluralism. In the first part, the article points out the anthropologic dimension and critical premises of the comprehension of a religious tradition. In the second part, important contributions from Jewish-Christian tradition to an effective inter-religious dialogue are presented: the dialogical structure of divine revelation, the universality of God’s love, the promotion of human life as a central value, the necessary historical incarnation of God’s experience and the sacramental dimension of religion in face of the theological centrality of God’s Kingdom. Finally, Torres Queiruga’s category of ‘inreligionation’ is used to show the wonderful results of inter-religious dialogue. The post-classical (or post-Avicennan, post-Rāzian) genre of the “protocols for dialectical inquiry and disputation” (ādāb al-baḥth wa-l-munāẓara) has its more proximate origins in the famed Risāla of Shams al-Dīn al-Samarqandī (d. 722/1322). The greater part of his conceptions and methodology, however, consists in a streamlining and universalizing of the more strictly juristic dialectic (jadal / khilāf) of his teacher Burhān al-Dīn al-Nasafī (d. 687/1288); and this in turn draws on the highly logicized dialectic of Rukn al-Dīn al-ʿAmīdī (d. 615/1218) and (...) his teacher Raḍī al-Dīn al-Nīsābūrī (d. 617/1220). At the heart of methods in this lineage, and carried forward by al-Samarqandī into the universal ādāb al-baḥth, are three truth-preserving logical relationships critical to the truth-seeking enterprise of dialectic: entailment (talāzum / mulāzama), mutual negation or exclusion (tanāfin / munāfā), and causal concomitance (dawarān). The practical elaboration of these relations reveals a logic in action—a premodern dialogical logic for living disputation praxis. In fact, so critical were these to the dialectical enterprise that al-Samarqandī devoted a specialized treatise entirely to summarizing their defining features and rules, aptly naming it the ʿAyn al-Naẓar, or “Wellspring of Rational Investigation.” In this article, and drawing upon a recently published digital critical edition, I will present an analytical outline of these core logical relations as presented in the ʿAyn al-Naẓar. Then I will address a number of points of interest in this text, grouped under six themes: the potential for cross-disciplinary advancement; notions in discursive development; significant or uniquely contributive formulations; peculiarities of content; signs of an evolving, universalist agenda; and evidence that the ʿAyn al-Naẓar was designed as an aide-mémoire for the active disputant. I write here about how the visible and the invisible intertwine in painting: in theory and in praxis – primarily the praxis of my own painting. Philosophers are rarely asked to discuss, much less to show in public, what they do avocationally rather than professionally. I was drawn to the invitation of the Merleau-Ponty Circle to exhibit my painting and to talk about what I do when I am not writing or teaching philosophy. It has offered a rare chance to (...) catch up with myself – with the painter in me. I started in art long before I turned to philosophy. It was my first love and is still my intermittent passion. I have not hidden it from others altogether – I have shown my work in various group shows– but it would be more accurate to say that I have hidden it from myself. Talking about my art work in the context of this Circle comes at a very welcome moment in which I am in the process of determining where I shall go with the rest of my life as it shortens down: ars longa, vita brevis.J’écris ici à propos de la manière dont le visible et l’invisible s’entrelacent dans la peinture : en théorie et en pratique, prioritairement dans ma propre pratique de la peinture. In lieu of an abstract, here is a brief excerpt of the content:The Senecan Moment:Patronage and Philosophy in the Eighteenth CenturyEdward AndrewThis piece examines the place of patronage in eighteenth-century thought and specifically Diderot's analysis of Seneca's philosophy of the art of graceful giving and grateful receiving.1 Patronage, in Burke's definition, is "the tribute which opulence owes to genius."2 However, the patronage of thought has been rarely discussed by political theorists, and when mentioned favorably by thinkers such as Rousseau or (...) Burke, their accounts were not a central theme of their work. When patronage was a central theme, such as Johnson's letter to Lord Chesterfield or D'Alembert's Essai sur la société des gens de lettres et des grands, the practice is deprecated as obsolete.3 Thinkers who pride themselves on their independence of thought have usually been reluctant to declare their dependence upon patrons. However, most of the thinkers of the eighteenth century who preached intellectual independence depended upon royal or aristocratic patronage.4 Indeed, one might say that the ideal of intellectual [End Page 277] autonomy was forged in conditions of intellectual dependence on royal or aristocratic patrons.Francis Bacon wrote that "books, such as are worthy of the name of books, ought to have no other patrons but truth and reason." Yet Bacon's Advancement of Learning, dedicated to James I, is forthright in admitting that most scholars are not independently wealthy and thus must court the rich and must submit to the powerful. In the same work Bacon denounced Seneca as a "Trencher Philosopher" who served and supped well at a royal court.5 Seneca's De Benefiis, the classic text on patronage, was often translated into English and French in the seventeenth and eighteenth centuries6 ; and it was highly regarded by eighteenth-century thinkers, such as La Mettrie, Lagrange, Rousseau, D'Holbach, Hume, Smith, and Diderot.7 Since Rousseau shared Hume's admiration for Seneca, Hume concluded his account of Rousseau's ingratitude for Hume's patronage, Exposé succinct de la contestation que s'est élevée entre M. But, Diderot added, "there is one human being who is exempted from this pantomime. That is the philosopher who has nothing and asks for nothing."9 Diderot seems to celebrate the self-sufficient Cynic Diogenes who lived from nature: "Whom does the savage beg from? The earth, the animals and fishes, the trees and plants and roots and streams." But Diderot knew that Diogenes was not [End Page 278] one of Jean-Jacques Rousseau's self-sufficient savages; Diogenes lived by begging from his fellow citizens and taking what he needed from others—for all property is common for the wise man. Indeed, the honest Diogenes lived much as did the truth-telling sponge, Rameau. Moreover, Diderot agreed with Rameau's form of Epicureanism—not that money is the chief good but that money is essential for life's vital pleasures. Diderot wrote: "I am far from despising sensual pleasures. I have a palate too and it is tickled by a delicate wine or dish" as well as by beautiful women and drunken parties with his friends.10In short, Diogenes's life would not have satisfied Diderot. The more useful model of the philosophic life was the immensely rich Stoic, Seneca. Alan William Raitt, a Fellow of the British Academy, went up to Magdalen College at the University of Oxford from King Edward's Grammar School in Morpeth, in 1948. He progressed from being an undergraduate there to graduate student, Fellow by Examination, Fellow, Tutor, and Senior Tutor, as well as serving the college as a distinguished Vice-President from 1983 to 1985. Raitt had by then already been named in 1976 Special Lecturer in French Literature for the university and, three years (...) later, University Reader. In 1992 he received the accolade of an ad hominem Chair. Raitt had a gift for friendship; one of his greatest friends was Pierre Castex. His reputation as an international authority on nineteenth-century French literature is second to none. Unlike some British and American scholars, Raitt is widely read and admired by the French themselves, and his name figures prominently in all bibliographies devoted to Villiers de l'Isle-Adam and Gustave Flaubert. Despite his many commitments, both in Oxford and in the sphere of French studies generally, he remained a consistently prolific scholar. (shrink)
